摘要:
目的探讨热籽感应加温对荷瘤小鼠的治疗效果。方法50只接种B16黑色素瘤的C57/BL小鼠随机分为两部分,第一部分7只,植入热籽后在磁场中加热20 min,分别对肿瘤中心、边缘和小鼠体温进行监测,加温结束后取下肿瘤组织,10%甲醛固定,组织学切片评估加热后肿瘤坏死情况。第二部分共43只小鼠,小鼠接种肿瘤后随机分为空白对照组(10只)、磁场对照组(10只)、热籽对照组(10只)、加温治疗组(13只),用于加温后小鼠生存期观察和瘤体大小动态观察。结果肿瘤中心位置加热5 min便可达到46 ℃,之后达到平台期,保持在50 ℃左右,而整个加温过程中小鼠体温无明显变化。加热后热籽周围肿瘤组织呈现凝固性坏死,加温治疗组60%(6/10)小鼠肿瘤生长受到抑制,生存期延长(P=00136,同空白对照组相比较)。结论热籽组合在小鼠肿瘤内可以取得较好的加热效果,单次较短时间的热籽感应加温治疗对荷瘤小鼠肿瘤生长有一定抑制作用,延长小鼠生存期。
Abstract:
ObjectiveTo evaluate the therapeutic effect of thermoseed inductive heating on tumorbearing mice. Methods Fifty C57/BL mice inoculated with B16 melanoma cells were randomly divided into two groups. Group 1 (7 mice) was implanted with thermoseed and heated in an alternating magnetic field for 20 minutes. Temperatures of the tumor center, tumor edge, and the rectum of the mice were monitored during the heating procedure. After the heating treatment, the mice were sacrificed, and the tumor was resected and fixed with 10% formalin. The necrosis of the tumor was evaluated histologically. The Group 2 (43 mice) was randomly divided into four subgroups: C group (control group, 10 mice), M group (magnetic control group, 10 mice), T group (thermoseed control group, 10 mice), and H group (heating treatment group, 13 mice). The tumor size and survival time of each group were observed continuously after the treatments. ResultsThe temperature of the tumor center reached 46 ℃ within 5 minutes, and then maintained at 50 ℃. No significant change of the rectal temperature was detected during the heating procedure. The tumor tissues around the thermoseeds showed coagulation necrosis after heating. In the H group, the tumor growth was inhibited in 6 of the 10 mice (60%). The survival time of the H group was significantly longer than that in the control group (P=0.0136).ConclusionsThermoseeds array can achieve promising heating effects in tumorbearing mice. Single and shorttime thermoseed inductive heating can inhibit the tumor growth and prolong the survival time of the mice.
