摘要:
目的探讨DNA损伤修复相关因子组蛋白H2AX和毛细血管扩张性共济失调突变因子(ataxia telangiectasia mutation,ATM)在宫颈癌组织中的表达水平及诊断意义。方法选取2016年1月~2017年7月我科宫颈癌30例(宫颈癌组),同时选取非宫颈癌18例为对照组,对宫颈癌组和对照组宫颈组织进行HE染色和免疫组化染色,在mRNA水平检测H2AX和ATM的表达情况,以磷酸化抗体检测磷酸化H2AX(γH2AX)与磷酸化ATM(phosphoataxia telangiectasia mutation,pATM)水平。结果荧光定量PCR分析宫颈癌组ATM mRNA为2.8546±0.8881,明显高于对照组1.0018±0.4980(t=9256,P=0.000);宫颈癌组H2AX mRNA为2.4312±0.8224,显著高于对照组0.7896±0.4791(t=8738,P=0.000)。γH2AX和pATM主要在细胞核中表达, γH2AX阳性率80.0%(24/30),显著高于对照组11.1%(2/18)(Z=-3.381,P=0000);宫颈癌组pATM阳性率83.3%(25/30),显著高于对照组16.7%(3/18)(Z=-4.031,P=0.000)。结论γH2AX和pATM在宫颈癌中高表达,作为DNA损伤修复因子对宫颈癌的早期诊断有一定的临床意义。
Abstract:
ObjectiveTo investigate the expression, activation, and significance on diagnosis of DNA damage repair factors H2AX and ataxia telangiectasia mutation (ATM) in cervical tissues from patients with cervical cancer. MethodsThirty patients with cervical cancer from January 2016 to July 2017 in our hospital were enrolled as study subjects (cervical cancer group). At the same time, 18 patients with noncervical cancer were selected as the control group. Both groups were given HE staining as well as immunohistochemical staining of the cervical tissues. The expressions of H2AX and ATM were detected at the mRNA level, and phosphorylated H2AX (γH2AX) and phosphoataxia telangiectasia mutation (pATM) were detected with phosphorylated antibody assays.ResultsThe realtime quantitative PCR showed the mRNA expression of ATM of (2.8546±0.8881) in the cervical cancer group, which was significantly higher than that in the control group (1.0018±0.4980, t=9.256, P=0.000). The mRNA expression of H2AX was (2.4312±0.8224) in the cervical cancer group, which was significantly higher than that in the control group (0.7896±0.4791, t=8.738, P=0.000). The γH2AX and pATM were mainly expressed in the nucleus of cancer cells. The positive rate of γH2AX in the cervical cancer group was 80.0% (24/30), significantly higher than that in the control group [11.1% (2/18), Z=-3.381, P=0000]. And the positive rate of pATM in the cervical cancer group was 83.3% (25/30), significantly higher than that in the control group [16.7% (3/18), Z=-4.031, P=0.000].ConclusionsThe γH2AX and pATM are highly expressed in cervical tissues from patients with cervical cancer. As the DNA damage repair factors, the γH2AX and pATM might have certain clinical significance in early diagnosis of cervical cancer.
