摘要:
目的通过体外模拟气腹环境,观察二氧化碳(CO2)和氦气(He)在不同压力下对胃癌细胞MKN45增殖、凋亡的影响。方法实验分为对照组、CO2组和He组,其中CO2组和He组又按照压力不同分为0、5、10、15 mm Hg组(作用时间均为4 h)。细胞在密闭培养箱内经不同压力CO2或He作用4 h后用血气分析仪检测培养液pH值;MTT法检测细胞增殖情况;流式细胞仪观察细胞凋亡指数变化;Annexin VFITC/PI双标染色、流式细胞仪测定凋亡细胞比例。结果处理结束后即刻检测,CO2组细胞培养液呈弱酸性,He组细胞培养液呈弱碱性。MTT法检测细胞增殖变化,CO2组0、5、10 mm Hg组OD值与对照组比较无明显差异(P>0.05),15 mm Hg组OD值与对照组比较在前3天无明显差异(P>0.05),在4~7天OD值明显低于对照组(P<0.01)。而在He组,不同压力组细胞增殖活性与对照组比较无明显差异(P>0.05)。在CO2环境下,0、5 mm Hg组细胞凋亡指数与对照组比较无明显差异(P>0.05),而10、15 mm Hg组明显大于正常对照组(P<0.01)。0、5 mm Hg组细胞凋亡比例与对照组比较无显著差异(P>0.05),而10、15 mm Hg组细胞凋亡比例明显大于对照组(P<0.01)。在He环境下0、5、10、15 mm Hg组细胞凋亡指数和凋亡比例较对照组均无明显差异(P>0.05)。结论在临床常用压力范围内(≤10 mm Hg),CO2对胃癌细胞增殖、凋亡无明显影响,而在15 mm Hg压力下,CO2抑制细胞的增殖。
Abstract:
ObjectiveTo investigate the effects of CO2 and helium insufflation administered at different pressures on the proliferation and apoptosis of cultured human gastric cancer cells, MKN45.MethodsThe gastric cancer cells MKN45 were divided into control, CO2, and helium groups. In CO2 and helium groups, the cells were exposed to CO2 or helium at different pressures (0, 5, 10, and 15 mm Hg) for 4 hours. And then pH of each culture media in the three groups was analyzed, the proliferation of the cells was detected by MTT assay, and the apoptotic index (AI) of each group was determined by flow cytometry. The cells were double labeled by AnnexinⅤFITC/PI staining, and the proportion of apoptotic cells in each group was tested by flow cytometry.ResultsThe cell media was weak acid in the CO2 group, and was weak basic in the helium group. MTT assay showed that the OD value of the 0, 5, and 10 mm Hg subgroups in the CO2 group was not significantly different from that in the control (P>0.05). Whereas, in the 15 mm Hg subgroup of the CO2 group, the OD value was similar to that of the control during the first 3 days (P>0.05), but decreased significantly from 4 to 7 days (P<0.01). The OD value of each subgroup of the helium group was similar to that of the control (P>0.05). In the CO2 group, the AI and proportion of apoptotic cells of the 0 and 5 mm Hg subgroups were not significantly different from those of the control (both P>0.05), while those of the 10 and 15 mm Hg subgroups were significantly higher than that of the control (both P<0.01). The AI and proportion of apoptotic cells in the helium group were similar to those of the control (both P>0.05). ConclusionsA pressure of CO2 ≤10 mm Hg do not influence the proliferation and apoptosis of gastric cancer MKN45 cells. When the pressure reach 15 mm Hg, the proliferation of the cells is inhibited.
